It can take a strong constitution.

After a while I developed a pretty good system. Forget the rubber
plugs. I just used a hot glue gun. Forget the little hole, I widened
it enough that the ink won't bubble up and shoot all over the place
when you squirt the ink in. The problem is that there is no way to get
the syringe into the hole without spilling a drop at the opening. So
when you press the plunger the ink bubble will splatter everywhere.

If you have the hole drilled clean enough the dollop of glue will get
sucked into the hole a little (specifically with canon cartridges as
there is a gravity/vacuum thing going on). When it comes time to
refill the empty cartridge, a knife or screwdriver will pry up the
glue and if the hole is smooth enough it will pop right out.

Use disposable plastic food gloves. When filling the cartridge, plug
the egress with a finger so the ink doesn't drain right out. You may
think that you're running out of fingers, but not really. You're just
holding the cartridge in a way that one of the fingers just happens to
plug the drain hole. Don't release it until the glue is in place. If
you time it right and the glue is just starting to congeal, remove
your finger from the drain hole and the glue will suck into the fill
hole just a little which provides a better seal, but not so much that
you can get it out next time.

The plastic food gloves can be washed off easily enough under a
faucet. I can usually do an entire set of inks without throwing away
the gloves. Make sure you have a syringe for each color. Do all of
them that need filling at one time then wash them all out with warm
water. fill, squirt, fill, squirt, remove plunger, fill, squirt, etc.,
until totally clean. Store disassembled or fully retracted. Before
filling with ink, squirt it a few times to eject any remaining water
from last time. Using a permanent marker, label each syringe with the
color. Always use the same syringe for a specific color. Especially
Yellow!!!

It's all easier than it sounds.  :)
